A win would bring England a step closer to qualification for the 2015 FIFA Women’s World Cup which takes place in Canada.
England’s next qualifier is against Montenegro on Saturday 5 April at Brighton’s Amex Stadium.
Sampson said: "I am pleased to be taking the team back to Shrewsbury Town and using their excellent stadium.
"The club and local community have shown fantastic support to the England Women's team in the past and I am sure this will be the case again in May.
"I really hope our supporters will be out in force come May for what will be a tough match for us against a strong Ukraine team."
Shrewsbury last hosted England Women in 2010, when they took on Switzerland as part of a successful 2011 FIFA Women’s World Cup qualifying campaign.
The club has also staged England Men’s U19, U20 and Under-21 internationals, including the Under-21s' Euro qualifier against San Marino in November last year.
Jayne Bebb, Shrewsbury Town club secretary, added: "It is a great honour to once again be able to welcome the England Women’s team to the Greenhous Meadow.
"Since their first visit back in November 2007 we have built up an excellent relationship with the team and The FA.
"It is great to have been asked to host this important game against the Ukraine and hopefully we can play our part in helping the team reach Canada 2015.
"It is an exciting time for women’s football in the country and to be able to help stage this game is another big boost for Shrewsbury Town Football Club."
England travel to Larnaca later this week to defend their Cyprus Cup title.
They will face Italy (on 5 March), Finland (7 March) and Canada (10 March) in the group stages of the annual friendly tournament.
